| LY354740, a synthetic agonist, blocks schizophrenia induced by the PCP model. This substance also prevents short-term memory impairment in schizophrenia. Low levels of glutamate play a role in the development of both positive and negative symptoms in schizophrenia. Lack of glutamate in the PFC and hippocampus causes an increase in dopaminergic activity by eliminating the braking effect on NMDA in the mesolimbic pathway. The DA increase in these pathways results in positive symptoms. |
